  • 神經科學領域的一個基礎問題就是網絡中的神經元如何互相溝通。為解決這個疑問,我們實驗室結合數種技術來研究相關的分子與細胞機制,包括 DNA 克隆與點突變、培養系統 ( 哺乳動物細胞株與初級組織培養 ) 、轉染技術 ( 活體內與體外電穿孔 ) 、免疫螢光化學、單細胞反轉錄定量聚合酉每連鎖反應、電生理記錄 ( 單一囊泡氧化電流測定 、 全細胞電壓或電流固定記錄 ) 以及活細胞即時影像記錄 ( 測量鈣離子動態變化、囊泡活動、以及螢光共振能量轉移顯影 ) 等。利用這些技術,我們以三個方向來探討神經傳導以及神經網絡發育的機制。 

  • 一:鈣離子調控的胞吐作用為神經傳導物質釋放的機制,我們以神經內分泌細胞株 (PC12 cells) 作為模式系統,來研究特定蛋白質其構造如何控制胞吐作用。透過氧化電流記錄的方法,我們可以解析單一囊泡釋放的各階段過程,我們的研究聚焦於胞吐作用中一個極為重要的動態中間點─”融合孔”,探討不同的蛋白如何藉由不同的訊息傳遞路徑以改變傳導物質釋放的過程,進而造成神經傳導物質釋放的多樣性。 

    二:我們將細胞株得到的結果應用於神經元中,以進一步探討中樞神經系統內傳導物質釋放的機制。首先,我們建立了迅速有效的方法,將目標基因送入初級培養的神經組織中。其次,在成體動物中,我們實驗室建立了一套將基因送入成鼠腦部的活體電穿孔技術,藉由操作特定腦區相關的蛋白,研究傳導物質如何在軸突或細胞本體釋放的分子機制。 

    三:我們使用發育中大鼠的視網膜作為研究神經網絡發育的系統。在特定的關鍵時期,週期性發生的模式化自發性放電現象會傳遞至一群發育中的神經元 ﹝例如發育中的視網膜波﹞,其功能在建構正確的神經網絡。過去的研究指出,自發性放電現象的特殊模式能夠引發發育網絡內 cAMP/PKA 的訊息傳遞,顯示視網膜波可以影響與基因表現相關的生化訊息路徑。我們於是利用單細胞反轉錄定量聚合酉每連鎖反應以偵測細胞內基因表現的改變,研究在細胞層級為什麼視網膜波可引起神經元連結的長效性改變,藉此影響神經網絡的建立。我們最終的目標是解析視網膜波的模式如何蘊含修飾神經網絡的訊息,使得視網膜能夠與大腦目標產生正確的連結。
